    Pharmacodynamics

    Methylcobalamin is a form of vitamin B 12 which helps in the production of myelin, a substance that protects nerve fibers and rejuvenates damaged nerve cells.

    Pregabalin is an alpha 2 delta ligand which decreases the pain by modulating calcium channel activity of the nerve cells. Together, they relieve neuropathic pain

    Absorption: Bind to intrinsic factor, a glycoprotein secreted by the gastric mucosa, and are then actively absorbed from the gastrointestinal tract.

     

        Absorption: Rapidly absorbed in GIT,

        Cmax= within 1.5 hr,

        Bioavailability= >90 %

     

    Distribution: Extensively bound to specific plasma proteins called transcobalamins. Diffuses across the placenta and also appears in breast milk.

      Distribution: Not bound to plasma proteins.Vss:         0.56L/kg.

     

    Elimination: Excreted in the bile, and undergoes extensive enterohepatic recycling; part of a dose is excreted in the urine, most of it in the first 8 hours; urinary excretion.

      Metabolism: Less than 2 % is metabolized

      Elimination:About 98% is excreted via urine as       unchanged drug

    Precaution

    Mecobalamin + Pregabalin should not be prescribed to individuals who are allergic to Pregabalin and vitamin B12 supplements
