    BRAND COMPOSITION DESCRIPTION
    TICLOX

    Each capsule contains:

    Cloxacillin Sodium BP eq. to Cloxacillin                500mg

    White to slightly yellowish blend filled in '0' size plain hard gelatin capsule shell having yellow body and green cap.

    PATIENT INFORMATION

    Administration:

    • TICLOX should be taken on empty stomach (1 hr before or 2hr after meal)
    • Do not break, chew, open, or crush the capsules. Swallow them whole.
    • Complete the full course of medication as prescribed even if the symptoms disappear after few days.

    Food interaction: Delayed absorption in the presence of food; Do not drink juice or carbonated beverage (soda) with TICLOX as it will decrease the effectiveness of the drug; Alcohol may irritate the stomach if taken with TICLOX.

    Pregnancy: Category B

    Nursing Mother: It is not known whether TICLOX passes into breast milk.

    Storage: Store in dry place below 25°C; Keep out of reach of children.

    Missed dose: Take the missed dose as soon as you remember; Skip the missed dose if it is almost time for your next scheduled dose; Do not take extra medicine to make up the missed dose.

     

    PHARMACOLOGY

    Pharmacotherapeutic Group: β-lactam antibiotic; Penicillin

     Pharmacodynamic properties: TICLOX (Cloxacillin) binds to penicillin-binding protein (PBPs) and inhibits the transpeptidation step of peptidoglycan synthesis in bacterial cell wall, thus inhibiting biosynthesis and arresting cell wall assembly. This causes formation of a defective cell wall that is osmotically unstable, resulting in bacterial cell death.

     Pharmacokinetic properties:

    Absorption: Incompletely absorbed from the GI tract with peak plasma concentrations after 1-2 hr; Oral bioavailability: 50%

    Distribution: Pleural and synovial fluids and bone (therapeutic concentrations), CSF (small amounts except when the meninges are inflamed; crosses the placenta and enters breast milk. Protein-binding: 94%

    Metabolism: Minimal metabolism.

    Excretion: Via the urine by glomerular filtration and renal tubular secretion (35% of an oral dose); via the bile (Up to 10%). Not removed by dialysis; Elimination half-life: 0.5-1 hr.

     DOSE

    250-500mg 4 times daily.

     CONTRAINDICATIONS

    Hypersensitivity to penicillins.

     SPECIAL PRECAUTIONS

    Renal impairment; Pregnancy and lactation; Porphyria; Cephalosporin hypersensitivity.

     ADVERSE DRUG REACTIONS

    Neutropenia, agranulocytosis; GI upsets; rash. Sore mouth or tongue; Black hairy tongue. Potentially Fatal: Neuromuscular hypersensitivity; pseudomembranous colitis; anaphylaxis.

     DRUG INTERACTIONS

    Coadministration of probenecid or disulfiram may result in higher cloxacillin concentration; Chloramphenicol and tetracycline antagonise bactericidal effect of penicillins; Cloxacillin increases the effect of methotrexate and side effect of allopurinol (may cause rashes); Cloxacillin may decrease the effectiveness of birth control pills.

    Potentially Fatal: Increased hypoprothrombinaemic effects of oral anticoagulants.
